Liquid Handling at the HTS @ The Nucleus

New HTS Robotic System in Sarafan ChEM-H W008

Liquid Handling Instrument iLab Schedules: Multidrop, Wellmate, PlateWasherVelocity11 VPrep, Agilent Bravo System and/or Plate Sealer.

The High-Throughput Screening @ The Nucleus has the ability to run 384- and 1536-well microplate based small molecule screens in a fully automated mode with the Thermo Automated Screening System which includes a Beckman Echo 655 acoustic dispenser. For siRNA screens we have an Agilent Bravo system . We also have stand alone instrumentation with stackers for running screens in "workstation" mode.

Thermo Integrated System

The Beckman Echo 655 acoustic dispenser is able to transfer compounds in 2.5 nL amounts from 384-well and 1536-well Echo compound plates to 96-, 384-, or 1536-well assay plates. The Echo is capable of adding compounds to any destination well from any source well at any volume. This allows it to be used to perform “hit picking” tasks (the transfer of a single sample from any position in a microplate to any other microplate), and to be used for automated dose response assays. 

Spinnaker 3 robotic arm is integrated to theEcho 655. The system is equipped with six 40-microplate stackers/or 18 plate hotels, a barcode reader and a delidder. The system is capable of shuttling microplates to and from the Echo and other stations on the deck. The Spinnaker integrates two Thermo Multidrop Combi dispensers, a BlueCatBio Bluewasher platewasher/dispenser, a Gyger Certus Flex dispenser, and an Agilent Bravo liquid handler with a 96- or 384-tip head, with the Echo 655 to permit the unattended, seamless completion of complex experimental protocols. Cell-based assays are further supported by the addition of a Thermo Cytomat  10C automated CO2 incubator to the system. This modification enables the automated shuttling of microplates between a cell culture-compatible environment, to the the Echo, and reagent dispensers/washers, making it possible to automate an entire cell-based assay. Finally, the robot is integrated to a BMG CLARIOstarPlus multimode plate reader. This allows for full automation of most assays.
