For automated large object sorting (10-200 um) and analysis we are utilizing the Union Biometrica BioSorter (datasheet, Brochure, User Manual, Quick Reference Guide) integrated to a Thermo Orbitor RS2 Microplate Mover (specs, Brochure, User Manual) for sorting to up to 200 microplates. (Automation on this system requires at least 1 month lead time as the automated robotic system is currently offline).

Union Biometric BioSorter
The BioSorter is a multi-range large particle flow cytometer (FACs) capable of sorting 10-1500 mircon objects to 6-, 12-, 24-, 48-, 96- or 384-well microplates. The BioSorter uses interchangeable Fluidics and Optics Core Assemblies (FOCA) to be able to cover the 10-1500 micron range, alhtough our current system only has the 250 um FOCA that can sort objects 10-200 microns. The 250 um FOCA is optimized for 20-150 micron objects such as C. elegans (i.e. worms, nematodes), and large cells (duct cells, hepatocytes, cardiomyocytes, and various stem cells), as well as beads in the 10-200 micron range. The BioSorter is then able to analyze the properties of the samples through the FlowPilot III software. Our current BioSorter configuration has both the standard 488nm excitation laser and the 561 nm laser permiting the detection of both GFP, YFP, and mCherry as well as other similar fluorescent proteins or fluorophores.
Automated RT PCR
We have also integrated the SFGF's Applied Biosystems (now part of Thermo) QuantStudio 12K Flex real-time PCR system (Schedule, Brochure) to the Thermo Orbitor RS2 to enable automation for such applications as qPCR and Protein Melt high-throughput screens in 384-well format.
