Anti-mouse mucosal vascular addressin, MAdCAM-1. Rat IgG2A. Recognizes the N-terminal Ig domain involved in a4b7 integrin binding. MAdCAM-1 is an Ig family member preferentially expressed in mucosal lymphoid tissues and lamina propria. It is a ligand for the lymphocyte mucosal homing receptor a4b7, and can also be decorated on high endothelial venules by L-selectin-binding carbohydrate determinants.

Rat IgG2a monoclonal antibody MECA-89 binds the second Ig domain of the mucosal vascular addressin, MAdCAM-1. Unlike MECA-367, MECA-89 does not block a4b7-dependent binding in vitro (although apparently through redistribution it depresses MAdCAM-1 function as an L-selectin ligand in Peyer’s patch-HEV in vivo). MAdCAM-1 is an Ig family member preferentially expressed in mucosal lymphoid tissues and lamina propria. It is a predominant ligand for the lymphocyte mucosal homing receptor a4b7

Induces a4b7-dependent lymphocyte aggregation, but inhibits other a4b7-mediated lymphocyte adhesion events including binding to MAdCAM-1 and a4b7 -dependent VCAM-1 and fibronectin binding. Useful in flow cytometry with sensitive second-stage reagents. Ineffective in immunohistology.

Anti-peripheral lymph node addressin (PNAd)-associated carbohydrate epitope. A rat IgM antibody that binds to a sulfate- and fucose-dependent carbohydrate epitope associated with L-selectin-binding carbohydrate ligands of high endothelial venules (HEV) in mouse lymphoid tissues and in human lymphoid tissues and sites of chronic inflammation. The antibody inhibits L-selectin-dependent lymphocyte homing to lymph nodes, and blocks in vitro binding to HEV (high endothelial venules) through L-selectin. The MECA-79 epitope decorates all known human and mouse HEV carbohydrate ligands for L-selectin including but not limited to CD34, glyCAM-1, and MAdCAM-1. The antibody reacts with HEV in numerous species (not rat).

An unusual mouse IgG2a anti-CD44 MAb which recognizes an extracellular sequence in the stalk of CD44, proximal to the hyaluronate (HA) binding globular domain but distal to the site of variant exon insertion (Ge and Butcher, unpublished). Hermes-3 blocks HA-independent lymphocyte binding to mucosal (appendix, Peyer’s patch) high endothelial venules (HEV) in vitro, by a mechanism which remains unknown. The antibody works well in paraffin section immunohistology.
