Mucosal Vascular Addressin (MAdCAM-1), mouse:

MECA-367 (HB9478): ATCC Number: HB-9478 Name: MECA-367

Anti-mouse mucosal vascular addressin, MAdCAM-1. Rat IgG2A. Recognizes the N-terminal Ig domain involved in a4b7 integrin binding. MAdCAM-1 is an Ig family member preferentially expressed in mucosal lymphoid tissues and lamina propria. It is a ligand for the lymphocyte mucosal homing receptor a4b7, and can also be decorated on high endothelial venules by L-selectin-binding carbohydrate determinants.

MECA-89: ATCC Number: HB-292 Name: MECA-89

Rat IgG2a monoclonal antibody MECA-89 binds the second Ig domain of the mucosal vascular addressin, MAdCAM-1. Unlike MECA-367, MECA-89 does not block a4b7-dependent binding in vitro (although apparently through redistribution it depresses MAdCAM-1 function as an L-selectin ligand in Peyer’s patch-HEV in vivo). MAdCAM-1 is an Ig family member preferentially expressed in mucosal lymphoid tissues and lamina propria. It is a predominant ligand for the lymphocyte mucosal homing receptor a4b7

ß7 Integrin Chain (mouse and human):

FIB504.64 ATCC Number: HB-293 Name: FIB504.64 and FIB21: ATCC Number: HB-295 Name: FIB21

Rat IgG2a antibodies to human and mouse ß7 integrin chain. FIB504 blocks lymphocyte binding to MAdCAM-1, a4b7 binding to VCAM-1 and fibronectin, and aeß7-dependent binding to intestinal epithelial cells. FIB21 also blocks, albeit less well, and is good for immunofluorescence and histology.

a4b7 heterodimer (Peyer’s patch homing receptor) in mouse.

DATK-32: ATCC Number: HB-294 Name: DATK32

Rat IgG2a monoclonal antibody specific for the mouse a4b7 integrin heterodimer. Induces a4b7-dependent lymphocyte aggregation, but inhibits other a4b7-mediated lymphocyte adhesion events including binding to MAdCAM-1 and a4b7 -dependent VCAM-1 and fibronectin binding. Useful in flow cytometry with sensitive second-stage reagents. Ineffective in immunohistology.

Peripheral lymph node addressin (high endothelial venule ligands for L-selectin).

MECA-79 (HB9479): ATCC Number: HB-9479 Name: MECA-79

Anti-peripheral lymph node addressin (PNAd)-associated carbohydrate epitope. A rat IgM antibody that binds to a sulfate- and fucose-dependent carbohydrate epitope associated with L-selectin-binding carbohydrate ligands of high endothelial venules (HEV) in mouse lymphoid tissues and in human lymphoid tissues and sites of chronic inflammation. The antibody inhibits L-selectin-dependent lymphocyte homing to lymph nodes, and blocks in vitro binding to HEV (high endothelial venules) through L-selectin. The MECA-79 epitope decorates all known human and mouse HEV carbohydrate ligands for L-selectin including but not limited to CD34, glyCAM-1, and MAdCAM-1. The antibody reacts with HEV in numerous species (not rat).

Cutaneous lymphocyte antigen (CLA), the skin lymphocyte homing receptor; and E-selectin binding glycotopes on myeloid cells and HEV.

HECA-452 (HB11485): ATCC Number: HB-11485 Name: HECA-452

Rat IgM antibody to the human lymphocyte homing receptor CLA, the cutaneous lymphocyte antigen. HECA-452 defines skin-homing memory T cells, binding to the T cell carbohydrate ligand for vascular E-selectin. HECA-452 also recognizes sialyl Lewisx and sialyl Lewisa and other carbohydrate ligands for E-selectin in man. HECA-452 also recognizes E-selectin-binding epitopes of high endothelial venules in man.

CD44 (human), aka H-CAM, Pgp-1, the Underhill hyaluronate receptor:

Hermes-3 (HB9480) ATCC Number: HB-9480 Name: Hermes-3

An unusual mouse IgG2a anti-CD44 MAb which recognizes an extracellular sequence in the stalk of CD44, proximal to the hyaluronate (HA) binding globular domain but distal to the site of variant exon insertion (Ge and Butcher, unpublished). Hermes-3 blocks HA-independent lymphocyte binding to mucosal (appendix, Peyer’s patch) high endothelial venules (HEV) in vitro, by a mechanism which remains unknown. The antibody works well in paraffin section immunohistology.
