Research InterestMy research is focused on attaining a better understanding of the initiation, maintenance, and progression of lymphoid tumors, and their response to immunochemotherapy toward improving current treatment strategies. In this effort, I employ tools from functional genomics, computational biology, molecular genetics, and mouse models. I hope to apply this knowledge towards the design of clinical trials in the treatment of patients with lymphoma, leukemia, and myeloma.

In addition to his clinical duties in the division of Oncology, Dr. Clarke maintains a laboratory focused on two areas of research: i) the control of self-renewal of normal stem cells and their malignant counterparts; and ii) the identification and characterization of cancer stem cells. A central issue in stem cell biology is to understand the mechanisms that regulate self-renewa..

Research InterestDr. Fan is a physician scientist who studies how turning off oncogenes (cancer genes) can cause tumor regression in preclinical and clinical translational studies. Based on her findings, she has initiated clinical trials studying how targeted therapies affect cancer signals in kidney cancer and low grade lymphoma. In the laboratory, she uses new nanotechnology strategies for tumor diagnosis and treatment to define biomarkers for personalized therapy.

Research InterestMy laboratory investigates how oncogenes initiate and sustain tumorigenesis. I have developed model systems whereby I can conditionally activate oncogenes in normal human and mouse cells in tissue culture or in specific tissues of transgenic mice. In particular using the tetracycline regulatory system, I have generated a conditional model system for MYC-induced tumors. I have shown that cancers caused by the conditional over-expression of the MYC proto-oncogene regress with its inactivation.

Research InterestWe study the genetics of pancreatic islet cell differentiation using molecular, embryologic and genetic methods in several model systems, including mice, embryonic stem cells, and Drosophila. Our work suggests that critical factors required for islet development are also needed to maintain essential functions of the mature islet. Our knowledge of genetic and cellular pathways governing islet formation has allowed us to use stem cell lines to produce islet replacements in vitro.

Research InterestThe recruitment of key effector immune cells to the tumor microenvironment is critical for improving outcomes in cancer treatment. Our research has focused on evaluation of a novel leukocyte chemoattractant, chemerin. We have shown it can act as a tumor suppressive cytokine by marshaling the host immune system and skewing the number and ratio of key anti-tumor effector cells at the site of tumor. We are continuing our preclinical studies of chemerin, and plan to expand the general approach.

Research InterestWe are working to elucidate the biochemical and cell biological principles that govern signaling pathways that sit at the intersection between developmental biology and cancer. Our toolkit combines bulk biochemical techniques, such as cell-free reconstitution, with microscopy using novel optical probes to study the dynamics of signal propagation in cells. We strive to develop novel strategies for the manipulation of these pathways for cancer therapies and applications in regenerative medicine.
